ABSTRACT

AIM: To analyze the effect of full-femtosecond small incision lenticule extraction(SMILE)on the treatment of high myopia based on propensity score matching.METHODS: A total of 48 cases(48 eyes)of high myopia patients who underwent SMILE surgery in our hospital from May 2019 to May 2021 were selected as the observation group, and 48 cases(48 eyes)of high myopia patients who underwent FS-LASIK surgery were matched using propensity score matching as the control group. Follow up for 6mo after surgery, the changes in cylindrical, central corneal thickness, uncorrected visual acuity(UCVA), corneal endothelial cell related indicators [percentage of hexagonal endothelial cells(6A), coefficient of variation(CV)of endothelial cell area, central corneal endothelial cell density(ECD)] and corneal biomechanical indicators [simulated Goldman intraocular pressure(IOPg), corneal hysteresis(CH), corneal resistance factor(CRF), corneal compensated intraocular pressure(IOPcc)] between the two groups were compared, and the incidence of complications in both groups of patients was recorded.RESULTS: Both groups of patients showed significant improvements in cylindrical and UCVA at 3 and 6mo after surgery, as well as decreased central corneal thickness, corneal endothelial cells, and corneal biomechanics related indicators. The changes in the observation group were more significant(all P<0.05). During the follow-up period, there was no significant difference in the incidence of complications between the observation group and the control group(8% vs. 17%, P>0.05).CONCLUSION: SMILE has a definite effect on patients with high myopia and is helpful to improve visual acuity.

ABSTRACT

Objective:To investigate the difference of dose distribution between intensity-modulated photon radiotherapy (IMRT) and intensity-modulated proton radiotherapy (IMPT) in patients with non-small cell lung cancer.Methods:The clinical data of 8 patients with stage Ⅱ-Ⅲ non-small cell lung cancer who received radiotherapy in Ion Medical center of the First Affiliated Hospital of University of Science and Technology of China from November 2020 to April 2022 were retrospectively analyzed. IMRT and IMPT radiotherapy plans were created for each patient separately, the main evaluation indicators were targeted area dose distribution parameters [homogeneity index (HI), conformity index (CI) and the percent volume of volume wrapped by 95% and 100% of prescription dose profile in the targeted area (V 95% and V 100%)], and the average dose (D mean) to the organ at risk and the percent volume of a certain relative biological effect (RBE) dose exposure [D mean, V 5 Gy(RBE) and V 20 Gy(RBE) of ipsilateral lung, D mean, V 5 Gy(RBE) and V 20 Gy(RBE) of bilateral lung, D mean, V 30 Gy(RBE) and V 40 Gy(RBE) of heart, maximum dose (D max) of spinal cord, and D mean of esophageal]. Results:In comparison with IMRT, IMPT reduced the levels of dose parameters in bilateral lung, ipsilateral lung, spinal cord, esophagus, and heart with statistically significant differences (all P < 0.05), especially in D mean of bilateral lung [(4.1±1.8) Gy (RBE) vs. (6.9±1.9) Gy (RBE)], V 5 Gy(RBE) [(15.9±7.1) % vs. (28.5±8.6)%], V 20 Gy(RBE) [(7.4±3.5)% vs. (10.1±3.5)%], and D mean of ipsilateral lung [(9.1±4.5) Gy (RBE) vs. (11.9±3.3) Gy (RBE)], all decreased significantly (all P < 0.001), but the differences in the levels of targeted area dose distribution parameters between them were not statistically significant (all P > 0.05). Conclusions:For patients with non-small cell lung cancer, IMPT is superior to IMRT in the protection of bilateral lung, ipsilateral lung, spinal cord, esophagus and heart.

ABSTRACT

Objective@#To explore the relationship between parental self efficacy consistency with children s social development,and to provide a reference basis for promoting children s social development.@*Methods@#During September to October of 2019, cluster sampling method were used to select 905 children and their parents from 2 kindergarten (senior , mid and junior class) and 2 primary schools (grade one to grade three) in Bengbu. Children s social development and parental self efficacy were assessed by the Strengths and Difficulties Questionnaire(SDQ) and the parenting sense of competence Scale, respectively. Ordinal Logistic regression was used to analyze the relationship between parenting efficacy consistency with children s social development.@*Results@#Prevalence of emotional problems, conduct problems, hyperactivity attention inability,peer problems, and abnormal prosocial behtavior was 8.95%,6.30%, 18.01%, 14.03%,7.40% and 5.41%,respectively, which were negatively associated with parental self efficacy( P <0.01). Consistent parenting sense of competence, children s emotion, hyperactivity attention inability, moral behavior and prosocial behavior anomaly detection rate lowest, mother parenting self efficacy were higher than the father, children s enotion, conduct behavior, hyperactivity attention inability and prosocial behavior anomaly detection rate is highest, when the father parenting self efficacy was higher than that of mothers, Children s conduct behavior and hyperativity attention inability had the highest detection rate( Z =6.57, 7.58,7.25, 7.06, P <0.05). Children with higher maternal parenting self efficacy were more likely to develop emotional, conduct behavior, hyperactivity attention inability and prosocial behavior abnormalities, and children with higher father parenting self efficacy were more likely to develop conduct behavior and hyperactivity attention inability ( P <0.05).@*Conclusion@#Parental self efficacy and its consistency are related to child social development.It is suggested that parents should improve the parenting efficiency and the quality of companionship, optimize the family relations, and create a harmonious atmosphere.

ABSTRACT

Objective@#The relationship between outdoor temperature and blood pressure (BP) has been inconclusive. We analyzed data from a prospective cohort study in northwestern China to investigate the effect of outdoor temperature on BP and effect modification by season.@*Methods@#A total of 32,710 individuals who participated in both the baseline survey and the first follow-up in 2011-2015 were included in the study. A linear mixed-effect model and generalized additive mixed model (GAMM) were applied to estimate the association between outdoor temperature and BP after adjusting for confounding variables.@*Results@#The mean differences in systolic blood pressure (SBP) and diastolic blood pressure (DBP) between summer and winter were 3.5 mmHg and 2.75 mmHg, respectively. After adjusting for individual characteristics, meteorological factors and air pollutants, a significant increase in SBP and DBP was observed for lag 06 day and lag 04 day, a 0.28 mmHg (95% @*Conclusions@#This study demonstrated a significant negative association between outdoor temperature and BP in a high-altitude environment of northwest China. Moreover, BP showed a significant seasonal variation. The association between BP and temperature differed by season and individuals' demographic characteristics (age, gender, BMI), unhealthy behaviors (smoking and alcohol consumption), and chronic disease status (CVDs, hypertension, and diabetes).

ABSTRACT

Objective RegIV, which is the latest member of the regenerative gene family, has been found to be closely related to the development of various tumors, but rarely reported in invasive breast cancer. This article is intended to explore the expression levels of regenerative factor 4(regeneration islet-derived family, member 4, RegIV), and the correlation with clinicopathological features and prognosis in 4 molecular subtypes of breast cancer(LA, LB,HER-2+, TNBC). Methods Pathological data of 284 breast cancer patients from March 2011 to October 2017 in the department of pathology, Lianyungang first people’s hospital were collected. To analyze the expression of RegIV in different molecular subtypes (LA, LB, HER-2+, TNBC) and its relationship with clinicopathological features, we analyzed the expression level of RegIVmRNA in breast cancer and its prognosis of patients. Results The positive rates of Reg IV in Luminal A group, Luminal B group, HER-2 positive group and triple negative breast cancer were 52.11%(37/71), 50.00%(42/84), 65.22%(45/69)and 20.00%(12/60),which was statistically significant when comparing them in pairs(P<0.01). The expression of RegIV was associated with tumor diameter, lymph node metastasis, TNM stage(P<0.01). The recurrence risk of breast cancer patients with high RegIV expression was lower than those with low RegIV expression (HR=0.59,95%CI:0.50-0.69). Further analysis found that the recurrence risk of LA breast cancer patients with high RegIV expression (HR=0.63,95%CI:0.48-0.82), and so it was with LB(HR=0.56,95%CI:0.40-0.76) and TNBC(HR=0.51,95%CI:0.36-0.71). Conclusion RegIV can provide reference for molecular subtypes diagnosis and it may be one of the indicators to evaluate the prognosis of breast cancer patients.

ABSTRACT

@#【Objective】To assess the value of 18F-FDG PET/CT for imaging liver fibrosis.【Methods】12 male SD rats (170±10 g)were divided into model group(TAA group)and control group. In the model group,200 mg/kg thioacetamide dissolved in sterile saline was administered to rats by means of intraperitoneal injection twice a week for a total of 6 weeks. In the control group,rats were treated with the same volume of saline. At week 6 after injection,18F- FDG PET/CT imaging was performed on two groups and measurement of the liver 18F-FDG uptake in two groups was taken. After PET/ CT scans, all rats were sacrificed to observe anatomical morphological changes. Liver tissues were harvested for hematoxylin and eosin (H&E) staining,Masson staining and measurement of hydroxyproline levels. 【Results】 Liver anatomical morphology of the TAA-induced rats was roughness with brunt margins and coarse surfaces ,while control rats showed sharp margins and smooth surfaces. HE staining showed visible histological changes that hepatocyte and liver sinus area surrounded by plentiful recruited hepatocyte neutrophils in the model group. Masson staining showed that obvious proliferation of fibroblasts and deposition of collagen in liver tissues in the model group. Model group showed higher hydroxyproline content than that in the control group(P<0.001). The results of 18F- FDG imaging indicated that apparent liver radioactivity concentration in the model group. 18F-FDG uptake value of liver tissues in the model group was higher than that in the control group(P<0.001).【Conclusion】18F- FDG PET/CT imaging could be used for diagnosis of liver fibrosis noninvasively.

ABSTRACT

Objective To test the reliability and validity of Chinese version of Illness Cognition Questionnaire(ICQ)in women with breast cancer. Methods From October,2016 to May,2017,297 women with breast cancer were investigated with the Chinese version of the ICQ and Brief Illness Perception Questionnaire(B-IPQ).The reliability and validity of the Chinese version of ICQ were tested. Results The Cronbach's α of three subscales of ICQ(helplessness,acceptance and perceive benefits)were 0.855,0.857, 0.878,and the test-retest reliability of three subscales were 0.946,0.963,0.901(P<0.001).A confirmatory factor analysis was tested,χ2/df=2.500,SRMR=0.063,CFI=0.920.The factor load capacity of each subscales was 0.37 to 0.84 and the correlation of helplessness,acceptance and perceive benefits was-0.49 to 0.81(P<0.001).The correlation coefficient of criterion-related validity of the helplessness subscale of ICP with B-IPQ was 0.727(P<0.001). Conclusion The Chinese version of ICQ has good reliability and validity,which is a valuable research tool in clinical studies of psychological cognition of patients with breast cancer.

ABSTRACT

Objective·To study the expression of transcription factor BTB and CNC homology 2 (Bach2) in CD4+CD25+CD45RA-T ceils from patients with systemic lupus erythematosus (SLE) and its effect on cell function.Methods·The CD4+CD25+CD45RA T cells from active SLE patients and healthy volunteers were sorted by flow cytometry.The expression of Bach2 in CD4+CD25+CD45RA T cells was detected by fluorescence quantitative PCR and Western blotting.The correlation between the median flourscence indensity (MFI) of Bach2 in CD4+CD25+CD45RA-T cells and the disease activity index of SLE (SLEDAI) was analyzed.The MFI of Bach2 in IL-17+CD4+CD25+CD45RA-T ceils was compared with that in IL-17-CD4+CD25+CD45RA-T cells by flow cytometry.In Bach2 overexpression system,the expression of IL-17 in CD4+CD25+CD45RA T ceils was detected by flow cytometry and the concentration of IL-17 in the culture supernants was detected by ELISA.Results·The mRNA and protein expressions of Bach2 in CD4+CD25+CD45RA-T cells from SLE patients were significantly lower than those in healthy controls (P<0.01).There was a significant negative correlation between the MFI of Bach2 and SLEDAI (R2=0.433,P=-0.001) in patients with SLE.The expression of Bach2 in IL-17+CD4+CD25+CD45RA-T cells was significantly lower than that in IL-17-CD4+CD25+CD45RA-T cells (P=-0.013).When Bach2 was overexpressed,the percentage of CD4+CD25+CD45RA-T cells from SLE patients expressing inflammatory factor IL-17 decreased significantly (P=0.032) and the IL-17 concentration in cell culture supernatants markedly decreased (P=0.008).Conclusion·The expression of Bach2 in CD4+CD25+CD45RA-T cells from SLE patients decreases,and overexpression of Bach2 in the cells leads to the falling expression of IL-17.

ABSTRACT

Objective To observe the retinal microstructure changes in patients with rhegmatogenous retinal detachment following vitrectomy procedures and its correlation with the best corrected visual acuity (BCVA).Methods Patients who were diagnosed with rhegmatogenous retinal detachment and underwent vitrectomy were recruited in this study.BCVA and optical coherence tomography (OCT) examination were performed at 1 week,1 month,3 months and 6 months after operation for recording the neurosensory retinal thickness and outer nuclear layer thickness as well as observing the occurrence of ellipsoid zone,external limiting membrane rupture,macuiar edema,macular epiretinal membrane.Results A total of 32 eyes from 31 patients were followed up,and the BCVA became better as the follow-up prolonged.Both neurosensory retinal thickness and outer nuclear layer thickness thickened from 1 week [(122.75 ± 111.84) μm and (79.06 ±76.67) μm] to 3 months [(163.28 ± 133.10) μm and (108.66 ± 120.40)μm],and slightly thinned at 6 months [(159.50 ± 99.26) μm and (99.22 ± 74.89) μm].AS follow-up went by,the occurrence rate of ellipsoid zone and external limiting membrane rupture were 68.8% and 53.1%,65.6% and 46.9%,56.3% and 40.6%,50.0% and 37.5% at 1 week,1 month,3 months,6 months,respectively,and both variables were significantly correlation with the patients' BCVA.Conclusion The neurosensory retinal thickness and outer nuclear layer thickness in patients with rhegmatogenous retinal detachment became gradually thicker and the vision became better during follow-up.Meanwhile,the ellipsoid zone and external limiting membrane rupture are the key factors for patients' BCVA.

ABSTRACT

Objective To prepare anti-folic acid (FA) polyclonal antibody and develop a new non-balanced competing chemiluminescence analysis for clinical detection of FA.Methods Established the detection method by added FITC-FA-analogs and FAHRP-antibody in the light emitting plate,which coated with anti-FITC antibody,to form the immune response complex of FITC/antibody-FITC-FA-analogs/FA-antibody-HRP.Then methodology evaluation was performed to evaluate the method performance;and further compared the detecting results with non-FITC system detection system and Electrochemiluminescence system (Roche Elecsys 2010).Results The ELISA results showed that the prepared anti-FA antibodies can recognize serum FA specificly.The methodology evaluation indicated that the linear correlation coefficient of the standard curve was 0.990 0;the analytical sensitivity was 1.21 ng/mL;the range of linear detection was 1.21~ 38.80 ng/mL;The coefficient variability of intra-assay was <5 %,which was better than the results of non-FITC detection system;and the correlation coefficient was 0.908 1 compared with the Elecsys-2010 detection system.Conclusion The established chemiluminescence immunoassay for human serum FA has a good sensitivity and specificity,and suitable for clinical serum FA quantitativedetecting.

ABSTRACT

Objective To detect the expression of BRIT1 in cervical cancer tissues and cervical noncancer tissues ,and to analyze the differences between the two tissues .Methods The expression of BRIT1 mRNA and protein in cervical cancer tissues and the paired cervical noncancer tissues was evaluated by RT‐PCR and immmunohistochemistry .Its correlation with the clinicopathological parameters including age ,tumor types ,size ,tumor pathological grade and clinical stage was analyzed .Results RT‐PCR results showed that the BRIT1 mRNA level in cervical cancer tissues was significantly lower than that in the paired cervical noncancer tis‐sues ,the difference was statistically significant (P<0 .05) .The immmunohistochemistry results showed that the BRIT 1 protein ex‐pression level in 44 cases of 63 (69 .8% ) samples wa slower than that in the paired cervical noncancer tissues ,the difference was statistically significant(P<0 .05);In high pathological grades and high clinical stages ,the decrease of BRIT1 protein expression was more significant .Conclusion The difference of the BRIT1 expression between the cervical cancer tissues and cervical noncancer tis‐sues suggests that BRIT1 may play a certain role in the occurrence and development of cervical cancer .

ABSTRACT

<p><b>OBJECTIVE</b>To prepare antibodies (pAbs) against phosphorylated Y-box binding protein 1 (pYB-1), perform qualitative detection of the ascites/pYB-1 ratio in patients with hepatocellular carcinoma with pulmonary metastasis (HCC-PM), and assess the clinical significance of the ascites/pYB-1 ratio as a diagnostic biomarker for HCC-PM.</p><p><b>METHODS</b>Bioinformatic prediction and chemical synthesis was used to identify and generate the YB-1 polypeptide with phosphorylation at serine position 102 (KYLRSVGDG). Rabbits were immunized with the YB-1 polypeptide coupled to a carrier protein. Protein A affinity chromatography was used to prepare highly-purified pAbs.ELISA and SDS-PAGE were used to determine concentration and purity of the pAbs. A total of 109 ascites specimens were collected from patients (36 cases of HCC,44 cases HCC-PM, and 29 cases of liver cirrhosis) and concentrated to obtain the pYB-1. Western blotting was used to qualitatively detect pYB-1 in ascites. Regression analysis and receiver operating characteristic (ROC) curve analysis were used to assess the qualitative data.</p><p><b>RESULTS</b>The prepared pAbs had a concentration of more than or equal to 1:1 * 106 and high purity. The pAbs/YB-1S102 specifically recognized endogenous pYB-1S102. The pYB-1S102 detected in ascites specimens from patients with HCC and HCC-PM, and the positive rate of detection was 30.6% and 77.3% respectively (P < 0.01).The pYB-1S102 showed sensitivity of 77.3% and a accuracy rate of 73.8% for diagnosis of HCC-PM.</p><p><b>CONCLUSION</b>Detection of pYB-1S102 in ascites could be a useful biomarker for diagnosis and metastasis monitoring in patients with HCC.</p>

ABSTRACT

Objective:To investigate the mechanisms of paeoniflorin on T cells in Bovine Collagen-induced arthritis ( CIA) model.Methods:CIA were established with Bovine Type Ⅱ collagen in DBA/1J mice.From day 21,after divided into two groups at random,mice were treated with PBS or paeoniflorin and recorded arthritis scores every day;the inflammation of CIA was evaluated with HE staining of the joints;the proliferations of splenocytes and CD4+T cells stimulated either by CD3 antibody and CD28 antibody or CII were examined by 3 H-TdR incorporation assay;the ratio of Th1 and Th17 subsets in CD4+T cells were examined by flow cytometry;the concentration of IFN-γand IL-17 in the culture supernatants of splenocytes were detected by ELISA.Results: The administration of paeoniflorin alleviated the symptoms( P<0.05) and inhibited the inflammation response of CIA;paeoniflorin not only inhibited the pro-liferation of CD3/28 antibodies-stimulated splenocytes and CD4+T cells(P<0.05),but also inhibited the proliferation of CII-specific splenocytes and CII-specific CD4+T cells(P<0.01);paeoniflorin modulated the ratio of Th1 and Th17 subsets(P<0.01);paeoniflorin reduced the levels of IFN-γand IL-17 in the culture supernatant of splenocytes ( P<0.05 ).Conclusion: Paeoniflorin may alleviates clinical symptoms of CIA via inhibiting pathogenic T cell proliferation, balancing inflammatory T cell subsets and reducing pro-inflammatory cytokines.

Objective To study whether the correlation exists between 6 kinds of autoimmune antibodies and female infertility . Methods The protein microarray technique was adopted to detect serum antibodies of anti-sperm antibodies(ASAb) ,anti-endome-trial(AEmAb) ,anti-zona pellucida(AZpAb) ,anti-ovarian(AVoAb) ,anti-hCG(AhCG Ab) and anti-trophoblastic(ATAb) in 140 ca-ses of female infertility and the correlation between these 6 kinds of antibody with female infertility was evaluated .Results Com-pared with the control group ,except AvoAb ,the positive rates of other 5 kinds of antibody had statistically significant differences (P0 .05) .The positive rate of ANA had no statistical difference between the infertility group and the control group (P>0 .05) ,but the titers of ANA in the experimenter were ≥1∶100(6/7) and most of karyotypes were nuclear coarse (5/7) ,which was differed from the low positive titer in the control group .The positive rate of ANA had statistical difference between the infertility related antibody positive and the infertility related antibody negative in the infertility patients .Conclusion ASAb ,AZpAb AEmAb , AhCGAb and ATAb are related with female sterility .The combined detection of these 6 kinds of antibody can significantly increase the detection sensitivity .

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the preventive effects and possible underlying mechanism of different extracts of Kanggushu () on osteoporosis in ovariectomized rats.</p><p><b>METHODS</b>One hundred and sixtyfive female SD rats were divided into 11 groups: control, sham, model, Xianling Gubao Capsule (), nilestriol, Kanggushu aqueous extract high-, medium-, and low-dose and suet extract high-, medium-, and low-dose groups. The osteoporosis model was made by ovariectomizing the rats. The latter 8 groups were administered intragastricly with Xianling Gubao Capsule, nilestriol, Kanggushu aqueous extract and suet extract for 12 weeks, respectively, while the other 3 groups were administered orally saline. The whole body bone mineral density, bone mineral content, organ coefficient of uterus, serum estradiol and alkaline phosphatase contents, blood calcium, phosphorus, interleukin 6 and bone Gla-protein levels after treatment were monitored. Additionally, three-point bending test of femur, HE staining, and scanning electron microscope were performed to explore the pharmacodynamics and underlying mechanisms.</p><p><b>RESULTS</b>In comparison with ovariectomized rats of model group, Kanggushu aqueous extract high-dose resulted in an increased bone mineral density, bone mineral content and organ coefficient of uterus, improved estradiol level, and improved maximum load and structural stiffness (P<0.05 or P<0.01). Two-dimensional and three-dimensional trabecular structure was also observed under HE staining and scanning electron microscopy, and the number and thickness of trabecular bone in Kanggushu aqueous extract high-dose group was significantly increased compared to the model group, while the lipid droplets in bone marrow cavity were significantly less. However, there were no significant differences in blood calcium, total serum alkaline phosphatase and bone Gla protein among different treatment groups. Overall, the osteoprotective effects of Kanggushu aqueous extract were comparable to those of nilestriol and were significantly more effective than those of Xianling Gubao Capsule.</p><p><b>CONCLUSION</b>The preventive effects of Kanggushu aqueous extract might be partly due to the increased estradiol level, accelerated restoration of bone trabecular reticulate structure, and accordingly increased bone mineral density in osteoporosis rats.</p>

ABSTRACT

To study the genotypes of hepatitis B virus (HBV) in patients from Chongqing district and determine the prevalence of autoantibodies in these patients. HBV genotyping was carried out by restriction fragment length polymorphism analysis of venous blood serum samples from 252 chronic hepatitis B (CHB) patients and 25 healthy controls. Indirect immunofluorescent assay was used to detect autoantibodies, including antinuclear antibody, anti-mitochondrial antibody, anti-smooth muscle (SM) antibody, and anti-liver and kidney microsomal antibody. Immunospot assay was used to detect anti-ribonucleoprotein/anti-SM antibodies, anti-SS-A antibody, anti-SS-B antibody, anti-scl-70 antibody, and anti-Jo-1 antibody. Correlations between the production of autoantibodies and patient age and sex and various genetypes of HBV were analyzed by the Chi-squared test. The most frequent HBV genotype in CHB patients was B (67.3%), followed by genotype C (32.7%). Genotypes A, D, E, F, G and H were not detected in any of the CHB patients. The positive rate of autoantibodies was higher in the CHB patients than in the healthy group (76.98% vs. 12.00%, X2 = 44.60, P less than 0.05). There was no significant differences in the autoantibodies profiles of CHB patients carrying the B or C genotypes ( X2 = 0.0016, P more than 0.05). The main HBV genotypes in CHB patients in the Chongqing district are B and C. Autoantibodies are prevalent among these CHB patients, and are correlated with patient age and course of liver disease but not HBV genotype or patient sex.

ABSTRACT

<p><b>BACKGROUND</b>Accumulating evidence indicates that both innate and adaptive mechanisms are responsible for the postnatal development of the mammalian visual cortex. Most of the studies, including gene expression analysis, were performed on the visual cortex during the critical period; few efforts were made to elucidate the molecular changes in the visual cortex during much earlier postnatal stages. The current study aimed to gain a general insight into the molecular mechanisms in the developmental process of the rat visual cortex using microarray to display the gene expression profiles of the visual cortex on postnatal days.</p><p><b>METHODS</b>All age-matched Sprague-Dawley rats in various groups including postnatal day 0 (P0, n = 20), day 10 (P10, n = 15), day 20 (P20, n = 15) and day 45 (P45, n = 10) were sacrificed respectively. Fresh visual cortex from the binocular area (Area 17) was dissected for extraction of total RNA for microarray analyses. Taking advantage of annotation information from the gene ontology and pathway database, the gene expression profiles were systematically and globally analyzed.</p><p><b>RESULTS</b>Of the 31 042 gene sequences represented on the rat expression microarray, more than 4000 of the transcripts significantly altered at days 45, 20 or 10 compared to day 0. The most obvious alteration of gene expression occurred in the first ten days of the postnatal period and the genomic activities of the visual cortex maintained a high level from birth to day 45. Compared to the gene expression at birth, there were 2630 changed transcripts that shared in three postnatal periods. The up-regulated genes in most signaling pathways were more than those of the down-regulated genes.</p><p><b>CONCLUSIONS</b>Analyzing gene expression patterns, we provide a detailed insight into the molecular organization of the developing visual cortex in the earlier postnatal rat. The most obvious alteration of gene expression in visual cortex occurred in the first ten days. Our data were a basis to identify new relevant candidate genes that control visual cortex development.</p>

ABSTRACT

Objective To study the (-)-epigallocatechin-3-gallate (EGCG) function on the proliferation of T cells derived from the peripheral blood and synovial fluid of rheumatoid arthritis (RA) and RA-related cytokine levels and the role of EGCG on RA synovial fibroblasts (FLS) proliferation was investigated. Methods ① Mononuclear cells from RA peripheral blood (30 cases) and synovial fluid (23 cases)were isolated. Blank group, negative control group, positive control methotrexate (MTX) group and therapeutic group with three different concentrations of EGCG were set up. Incorporated isotope 3H was used to test T cell proliferation from RA-PBMC and SFMC. ELISA assay was used to test cytokine (TNF-α, IFN-γ, IL-1, IL-6 and IL-17A) levels. ② MTT assay was used to test FLS proliferation from RA synovial tissue (8 cases).Results ① The CPM value of the high-dose group of EGCG in the peripheral blood and synovial fluid of RA patients was [ ( 15 136±2910), ( 11 587±3135 ) ], which was declined significantly than the control group (42856±2127) (P<0.01). The levels of TNF-α, IFN-γ, IL-1, IL-6 and IL-17A in the high-dose group of EGCG in the peripheral blood were [(321±13), (298±20), (132±12), (197±7), (59±8) pg/ml], which were decreased significantly than those of the control group [ (458±28), (505±26), (346±28), (405±25),(109±13) pg/ml ] (P<0.05 or P<0.01 ). The levels of TNF-o, IFN-γ, IL-1, IL-6 and IL-17A in the highdose group of EGCG in the synovial fluid were [(41.4±2.9), (182±16), (56.3±11.0), (34.2±1.9), (44±8)pg/ml ], which was decre-ased significantly than the control group [ ( 388.3± 19.3 ), (469±20), ( 104.2±17.8 ),( 114.5±4.8), ( 104±11 ) pg/ml] (P<0.05 or P<0.01 ). ② The level (A) of the high-dose group of EGCG in the FLS was (0.08±0.02), which was declined significantly than the blank group (0.27±0.04) (P<0.05).Conclusion ① In vitro EGCG can inhibit T cell proliferation from peripheral blood and synovial fluid of RA patients and the TNF-α, IFN-γ, IL-1, IL-6 and IL-17A secretion are decreased. ② In vitro EGCG can inhibit the proliferation of RA FLS.

ABSTRACT

Objective The therapeutic effect of epigallocatechin-3-gallate (EGCG) on the collageninduced arthritis model (CIA) was observed and its immunological mechanism was analyzed. Methods EGCG was administered to CIA mice and PBS was admitted as negative control. The severity of CIA was evaluated by clinical scores and histopathological assessment (H-E staining). Immunological mechanisms inv-suppressive effect on IL-17 secretion of CD4+T cells (EGCG group: 0.41%; PBS group: 4.05% ) and inhibitive activity of C Ⅱ -reactive splenocytes proliferation. There was statistical significant difference between IKB expression and down-regulate phosphorylated IKB expression in lymph node cells of CIA mice.Conclusion EGCG can significantly ameliorate the severity of CIA. The therapeutic mechanisms may be related to inhibition of C Ⅱ -reactive splenocyte proliferation and IL-17 secretion and via inhibiting the activity of NF-κB by inducing the expression of IKB and by suppressing the expression of phosphorylated IKB in CIA mice.

ABSTRACT

<p><b>OBJECTIVE</b>To demonstrate the vasodilatation activity of the coumarin-containing Angelica dahurica var. formosana and to further analyze active components in the herb extracts.</p><p><b>METHODS</b>(1) The vasodilatation effects induced by different extracts (cyclohexane, ethyl acetate, acetone, methanol, 95 % ethanol and water) of Angelica dahurica var. formosana on mouse thoracic aorta pre-contracted with phenylephrine were investigated. (2) The amount of imperatorin and isoimperatorin in each extract was measured by high-performance liquid chromatography. (3) The vasodilatation effects of imperatorin and isoimperatorin on mouse thoracic aorta were compared using the same in vitro method. (4) The vasodilatation mechanism of imperatorin in the mouse thoracic aorta pre-contracted with phenylephrine was studied using the methods of denuded endothelium, NG-nitro-L-arginine methylester (L-NAME, a nitric oxide synthase inhibitor), and propranolol.</p><p><b>RESULTS</b>(1) The cyclohexane and ethyl acetate extracts of Angelica dahurica var. formosana decreased the maximal response of phenylephrine-induced mouse thoracic aorta contraction dose-dependently, with 50% inhibiting concentration (IC(50)) values of 35.3+/-12.4 mg/L and 40.5+/-12.0 mg/L, respectively. The vasodilatation effect of imperatorin and isoimperatorin was dose-dependent. (2) The cyclohexane extract, showing the strongest vasodilatation effect, possessed the highest contents of imperatorin (4.09%) and isoimperatorin (0.27%, w/w). There was a correlation between the vasodilatation activity and the contents of imperatorin and isoimperatorin in the extracts. (3) The vasodilatation effect of imperatorin was about 4-fold stronger than that of isoimperatorin. (4) The vasodilatation effect of imperatorin was signifificantly attenuated to 24.88%+/-4.06% in the denuded endothelium group compared with the intact endothelium group. And 1 mmol/L L-NAME reduced the imperatorin-induced vasorelaxation by 32.18 %+/-11.29 %.</p><p><b>CONCLUSIONS</b>The principal effective component of Angelica dahurica var. Formosana was found to be imperatorin. Imperatorin-induced vasodilatation is at least partially regulated by nitric oxide, and has no correlation to beta-receptor.</p>
